In an interview for a regional, should you answer a question, alternate requirements, in 121 or 91 requirements. I would say 121 if you know the answer but should they tell you what they want? Thanks

Answer what you know. If you have never flown 121 then answer 91 or 135. You should know the rules you currently fly under. They can not expect you to know the rules that you want to fly under. That is what class is for.
