I'm curious what minimum requirements are for part 135 passenger-carrying operations. Can a 135 pilot carry passengers in an airplane approved for single pilot operation? What are the biggest obstacles to starting a part 135 operation (besides purchase and maintenance)??
PIC needs min 500TT for VFR operations, 1200TT for IFR.
Biggest headaches are getting the 135PIC qualified & the aircraft to be used 135 qualified plus getting the operations manual written & approved.

§ 135.243 Pilot in command qualifications.
(a) No certificate holder may use a person, nor may any person serve, as pilot in command in passenger-carrying operations --
(1) Of a turbojet airplane, of an airplane having a passenger-seat configuration, excluding each crewmember seat, of 10 seats or more, or of a multiengine airplane in a commuter operation as defined in part 119 of this chapter, unless that person holds an airline transport pilot certificate with appropriate category and class ratings and, if required, an appropriate type rating for that airplane.
So as you will see below, if you are engaged in Commuter Operations using a Multi-Engine Airplane, (other than turbojet powered), the aircraft may be single pilot certificated but you will need an ATP....which means 1500TT.
§ 119.3 Definitions
Commuter operation means any scheduled operation conducted by any person operating one of the following types of aircraft with a frequency of operations of at least five round trips per week on at least one route between two or more points according to the published flight schedules:
(1) Airplanes, other than turbojet powered airplanes, having a maximum passenger-seat configuration of 9 seats or less, excluding each crewmember seat, and a maximum payload capacity of 7,500 pounds or less...
The criteria mentioned in the previous post is certainly valid for some operations, but you should be aware of the differences.
Is it possible for a single person to operate a 135 on demand charter business?? I've been looking at the Regs and it appears that certian job positions must be filled. ie Chief of operations, chief of maintance etc...
There are several levels to the 135 Certificate. As a single entity/person you could be a single pilot operator or a single pilot-in-command operator (allows for SIC). Next level is a basic operator, which is limited to 5 aircraft and 5 PICs (I believe). Operations manual, training program, and the management personnel are not required for the single and single PIC operators. There will be some restrictions for those that other operators can do.
CVSFLY is right, what you are talking about would be defined as a single-pilot-in-command 135 air taxi operator. This would not be commuter flying as a few post before.
And I became a captain in a 135 outfit (multiple planes) flying a Lear with a little more than 1200TT, I had 1250 or something. Offcourse I needed the ATP in a turbojet to fly with PAX. WHich I got as soon as I got the hours.
So it would be possible especially since you are talkiing about being a single-pilot operation, just the big question will be "who is going to insure you, with only 1200TT".
